A Bayesian approach to estimate the accuracy of "in-house" ELISA assay to measure rabies antibodies from compulsory vaccinated dogs and cattle J. Venom. Anim. Toxins incl. Trop. Dis.
Nogueira,AHC; Silva,C; Gomes,DE; Rosa,ACG; Luvizotto,MCR; Cardoso,TC.
Rabies is a vaccine-preventable disease that causes acute encephalitis in mammals, and it is still a significant public health problem in numerous countries. Infected dogs represent the main vectors involved in human rabies. Additionally, cattle rearing close to geographic areas where vampire bats are found presents an important connection with rural epidemiology. We applied two "in-house" enzyme-linked immunosorbent assay (ELISA) methodologies, considered alternatives to measure antibodies from vaccinated dogs and cattle, without employing the gold standard approach. The ELISA assays were performed on individual serum samples taken from domestic adult dogs and cows compulsory vaccinated against rabies (147 urban dogs and 64 cows; n = 211). The sandwich...

Silva,Nylber Augusto da; Alves,Ângelo Giuseppe Chaves; Albuquerque,Ulysses Paulino de; Ramos,Marcelo Alves.
ABSTRACT We employed a biocultural approach to understanding the dynamics of knowledge ans use of natural resources associated in immaterial aspects of culture. We investigated whether factors such as sex, income, age, religion, occupation and time of participation in cultural practice influence the richness of species known and used by members of Cavalo Marinho, expression of the popular culture of the Brazilian Northeast that brings together theater, music and dance. We recorded a total of 111 ethnospecies (95 plants and 16 animals), based on information obtained from 56 informants. .There was a predominance of native plants and domestic animals in the knowledge of the participants in this cultural expression, although effective use is restricted to few...

A biodegradation study of forest biomass by Aspergillus niger F7: correlation between enzymatic activity, hydrolytic percentage and biodegradation index BJM
Sharma,Nivedita; Kaushal,Richa; Gupta,Rakesh; Kumar,Sanjeev.
Aspergillus niger F7 isolated from soil was found to be the potent producer of cellulase and xylanase. The residue of forest species Toona ciliata, Celtris australis, Cedrus deodara and Pinus roxburghii was selected as substrate for biodegradation study due to its easy availability and wide use in industry. It was subjected to alkali (sodium hydroxide) treatment for enhancing its degradation. Biodegradation of forest waste by hydrolytic enzymes (cellulase and xylanase) secreted by A. niger under solid state fermentation (SSF) was explored. SSF of pretreated forest biomass was found to be superior over untreated forest biomass. Highest extracellular enzyme activity of 2201±23.91 U/g by A. niger was shown in pretreated C. australis wood resulting in...

A biogeographical population genetics perspective of the colonization of cats in Latin America and temporal genetic changes in Brazilian cat populations Genet. Mol. Biol.
Ruiz-García,Manuel; Alvarez,Diana.
We used nine morphological genes to analyze cat populations from Mexico, the Dominican Republic, the Colombian, Brazilian and Peruvian Amazon, Bolivia and Brazil. Most populations were in Hardy-Weinberg equilibrium at the O locus. The highest allele frequencies so far detected at world level for alleles I (inhibitor) and L (long hair) were found at La Paz (Bolivia). The analyses revealed at least five cat gene pools in Latin America: These findings suggest that the current genetic distribution of cats in Latin America correlates with the colonization of the Americas during the XIV to XVIII centuries. Additionally, the cat populations of São Paulo, Rio de Janeiro and Manaus were sampled, to compare their 1996-2003 genetic profiles with those obtained in...

A biolistic process for in vitro gene transfer into chicken embryos BJMBR
Ribeiro,L.A.; Mariani,P.D.S.C.; Azevedo,J.L.; Rech,E.L.; Schmidt,G.S.; Coutinho,L.L..
Chicken embryos kept in culture medium were bombarded using a high helium gas pressure biolistic device. To optimize the factors that affect transformation efficiency, the lacZ gene under control of the human cytomegalovirus immediate early enhancer/promoter was used as a reporter gene. There was an inverse relationship between survival rate and transformation efficiency. The best conditions obtained for high embryo survival and high transformation efficiency were achieved with 800 psi helium gas pressure, 500 mmHg vacuum, gold particles, an 8 cm DNA-coated microparticle flying distance to the embryo and embryo placement 0.5 cm from the center of the particle dispersion cone. Under these conditions, transformation efficiency was 100%, survival rate 25% and...

A biomechanical study on proximal junctional kyphosis following long-segment posterior spinal fusion BJMBR
Zhu,Wen-Yi; Zang,Lei; Li,Jian; Guan,Li; Hai,Yong.
Posterior long-segment spinal fusion may lead to proximal junctional kyphosis (PJK). The present study sought to identify the appropriate fusion levels required in order to prevent PJK using finite element analysis. A finite element model was constructed based on the whole-spine computed tomography findings of a healthy adult. Nine commonly used posterior spinal fusion methods were selected. Stress on the annulus fibrosis fibers, the posterior ligamentous complex, and the vertebrae after various spinal fusions in the upright position were compared. This study was divided into two groups: non-fusion and fusion. In the former, the stress between the T10 and the upper thoracic vertebrae was higher. Comparing thoracic and lumbar segments in the fusion group,...

A bovine herpesvirus 5 recombinant defective in the thymidine kinase (TK) gene and a double mutant lacking TK and the glycoprotein E gene are fully attenuated for rabbits BJMBR
Silva,S.C.; Brum,M.C.S.; Weiblen,R.; Flores,E.F.; Chowdhury,S.I..
Bovine herpesvirus 5 (BoHV-5), the agent of herpetic meningoencephalitis in cattle, is an important pathogen of cattle in South America and several efforts have been made to produce safer and more effective vaccines. In the present study, we investigated in rabbits the virulence of three recombinant viruses constructed from a neurovirulent Brazilian BoHV-5 strain (SV507/99). The recombinants are defective in glycoprotein E (BoHV-5gEΔ), thymidine kinase (BoHV-5TKΔ) and both proteins (BoHV-5gEΔTKΔ). Rabbits inoculated with the parental virus (N = 8) developed neurological disease and died or were euthanized in extremis between days 7 and 13 post-infection (pi). Infectivity was detected in several areas of their brains. Three of 8 rabbits inoculated with the...

A bovine teat papilloma specimen harboring Deltapapillomavirus (BPV-1) and Xipapillomavirus (BPV-6) representatives BABT
Claus,Marlise Pompeo; Lunardi,Michele; Alfieri,Amauri Alcindo; Otonel,Rodrigo Alejandro Arellano; Ferracin,Lara Munique; Fungaro,Maria Helena Pelegrinelli; Alfieri,Alice Fernandes.
The common occurrence of multiple papillomavirus infections has been shown in several studies involving the human host. However, investigations with the aim of identifying mixed papillomavirus infections in cattle have been conducted only recently. In the current work we describe a co-infection with two different bovine papillomavirus (BPV) types that was identified in a bovine teat papilloma. The skin wart was obtained from a cow belonging to a Brazilian beef herd. A PCR assay was carried out with the FAP primer pair, which amplifies a partial segment of the L1 gene (approximately 478 bp), and the amplicon was submitted to direct sequencing. Because nucleotide sequences with satisfactory quality scores were not obtained, the amplicon was cloned and...

A box-model of metal flows through the Scheldt estuary (1981-1983 and 1992-1995) OMA
Baeyens, W.F.J.; Monteny, F.; Van Ryssen, R.; Leermakers, M..
The behaviour of Cu, Zn, Cd, and Pb in the Scheldt estuary was investigated in the periods 1981 to 1993 and 1995. The Hg behaviour was studied between 1992 and 1994. A box model was used to establish the relationship between the trace metal inputs and the fate of these pollutants in the Scheldt estuary. Annual budgets were calculated for particulate and dissolved trace metals. For Hg, seasonal budgets of the various species (inorganic Hg, methylmercury, Hg°) could also be calculated. These budgets allow an evaluation of the consistency of each individual flux or process, such as the inputs of the various metals into the North Sea, the sedimentation flux of particulate metals in the area of high turbidity, the evasional fluxes and the formation rates of...

A bradykinin-potentiating peptide (BPP-10c) from Bothrops jararaca induces changes in seminiferous tubules J. Venom. Anim. Toxins incl. Trop. Dis.
Gilio,Joyce M; Portaro,Fernanda CV; Borella,Maria I; Lameu,Claudiana; Camargo,Antonio CM; Alberto-Silva,Carlos.
Background The testis-specific isoform of angiotensin-converting enzyme (tACE) is exclusively expressed in germ cells during spermatogenesis. Although the exact role of tACE in male fertility is unknown, it clearly plays a critical function in spermatogenesis. The dipeptidase domain of tACE is identical to the C-terminal catalytic domain of somatic ACE (sACE). Bradykinin potentiating peptides (BPPs) from snake venoms are the first natural sACE inhibitors described and their structure-activity relationship studies were the basis for the development of antihypertensive drugs such as captopril. In recent years, it has been showed that a number of BPPs - including BPP-10c - are able to distinguish between the N- and C-active sites of sACE, what is not...

A brain microdialysis study on 5-HT release in freely moving rat lines selectively bred for differential 5-HT1A receptor function BJMBR
Gonzalez,L.E.; Parada,M.A.; Tucci,S.; Teneud,L.; Overstreet,D.H.; Hernandez,L..
Breeding for high and low hypothermic responses to systemic administration of a serotonin1A (5-HT1A) receptor agonist (8-hydroxy-2-(di-n-propylamino)tetralin, 8-OH-DPAT) has resulted in high DPAT-sensitive (HDS) and low DPAT-sensitive (LDS) lines of rats, respectively. These lines also differ in several behavioral measures associated with stress. In the present microdialysis study we observed that basal 5-HT concentrations in the prefrontal cortex and dorsal hippocampus did not differ significantly between HDS and LDS rats. Thus, behavioral differences between the HDS and LDS lines might not be attributed to differences in basal 5-HT release. However, both lines had lower basal levels of 5-HT release than their randomly bred control group (random...

A Brazilian Bacillus thuringiensis strain highly active to sugarcane borer Diatraea saccharalis (Lepidoptera: Crambidae) BJM
Gitahy,Patrícia de Medeiros; Souza,Marlene Teixeira de; Monnerat,Rose Gomes; Arrigoni,Enrico de Beni; Baldani,José Ivo.
The control of the major sugarcane pest, Diatraea saccharalis, is limited by the stem location of the caterpillar. As part of a long-term project towards the development of an alternative and efficient delivery system of Cry proteins to control the sugarcane borer, the current work describes the selection and characterization of a Brazilian B. thuringiensis strain with prominent activity towards D. saccharalis. Strain S76 was eleven-fold more active than the HD-1 Lepidoptera-standard strain, as estimated by the LC50 of 13.06 µg/L and 143.88 µg/L, respectively. We observed bipiramidal and cuboidal crystals similar to those found in other B. thuringiensis strains with entomopathogenic activity against Lepidoptera and Diptera. In addition, smaller and...

A Brazilian family with hereditary inclusion body myopathy associated with Paget disease of bone and frontotemporal dementia BJMBR
Fanganiello,R.D.; Kimonis,V.E.; Côrte,C.C.; Nitrini,R.; Passos-Bueno,M.R..
Inclusion body myopathy associated with Paget disease and frontotemporal dementia (IBMPFD) is a progressive and usually misdiagnosed autosomal dominant disorder. It is clinically characterized by a triad of features: proximal and distal myopathy, early onset Paget disease of bone (PDB), and frontotemporal dementia (FTD). It is caused by missense mutations in the valosin-containing protein (VCP) gene. We describe here the clinical and molecular findings of the first Brazilian family identified with IBMPFD. Progressive myopathy affecting the limb girdles was detected by clinical examination followed by muscle biopsy and creatine kinase measurement. PDB was suggested after anatomopathological bone examination and FTD was diagnosed by clinical,...
